Spoke/Comcast wheel compatibility question from a NEWB

I've tried searches and surfing the web and I'm more confused than not. I just bought a 1980 CB750C with comcast wheels, and I have a set of (year unknown) spoked 750 wheels that I want to use for a cafe' racer conversion. Both sets have rear drum brakes and front single discs. So far so good. I just don't know if these wheelsets are universally compatible or if there are differences that are not readily apparent.
One additional detail. The rear axles differ in one small way; the left end of the comcast wheel on the bike has a hex head so one could use a wrench to stabilize it while tightening the right side axle nut. The spoked one has a solid end "cap" with small holes that could utilize a screwdriver or similar to stabilize it.
Any help from the obviously knowledgeable bunch here would be greatly appreciated.
